Controlling gene expression with hydrogen peroxide 'switches'

Hydrogen peroxide doesn't just come in bottles from the drugstore ? the human body makes it as well. Now researchers from North Carolina State University have found a way to use naturally occurring hydrogen peroxide inside cells to switch on gene expression. Their method also serves as a highly sensitive hydrogen peroxide detector, which may help scientists determine the molecule's role in cellular health and disease.

In a normally functioning cell, hydrogen peroxide serves as a messenger, carrying signals through a cell in order to allow the cell to respond to external stimuli or events. Once the message is transmitted, the hydrogen peroxide diffuses and disappears.

"It's an ideal messenger compound, because it's small, fast, and doesn't linger," says chemist Alex Deiters. "It works by oxidizing, or modifying, certain amino acids in proteins, which affects the protein's function."

Deiters and graduate students Jeane Govan, Andrew McIver and Chad Riggsbee wanted to see if they could harness hydrogen peroxide's oxidizing property as a way to control gene expression, using the gene that gives fireflies their "glow," or luminescence, as a test case. They designed a molecule that was sensitive to hydrogen peroxide and enabled expression of the firefly luciferase gene in live mammalian cells. When hydrogen peroxide was present, the luciferase gene was expressed, causing the cell to glow.

The researchers' results appear online in Angewandte Chemie.

"The exciting aspects of this synthetic gene switch are that you can use it not only as a way to detect the presence of hydrogen peroxide within cells, in this case, by making cells containing hydrogen peroxide glow, but also to express any gene of interest in response to the presence of hydrogen peroxide," Deiters says. "In terms of detecting hydrogen peroxide, the system is at least an order of magnitude more sensitive than anything used previously."

Phelps back on top, beats Lochte in 200 IM

LONDON (AP) ? Michael Phelps added to his medal collection with his first individual gold of the London Games, and handed Ryan Lochte a double disappointment on his rival's final night in the pool.

Phelps set the tone right from the start Thursday to become the first male swimmer to win the same individual event at three straight Olympics, capturing the 200-meter individual medley for his 20th career medal ? and 16th gold. He touched in 1 minute, 54.27 seconds, just off his winning time in Beijing, but still good enough for gold.

Lochte settled for silver and Hungary's Laszlo Cseh took the bronze.

So a farewell games that started as a bit of a disappointment for Phelps is definitely looking up. He's now won two golds and two silvers in five races ? not up to his standards when he went 8-for-8 in China four years ago, but a fitting capper to a brilliant career that still has two more events to go.

In fact, as soon as Phelps finished off Lochte, he hopped out of the pool and headed to the nearby diving well to warm down, knowing he still had a semifinal of the 100 butterfly before the night was done. He was the top qualifier in that one, setting up a rematch in Friday's final against Milorad Cavic ? the outspoken Serbian who still seems to think he got to the wall first in Beijing but lost by a hundredth of a second.

Lochte had gone through the same warm-down routine just a few minutes earlier, trying to pull off an impressive double 31 minutes apart. He came up short in both races, fading to bronze in the 200 backstroke behind fellow American Tyler Clary, then touching after Phelps in the medley.

Phelps' reaction wasn't a water-pounding celebration, just a dazed smile and a definite look of relief. He seemed to be soaking it all in, relishing a gold of his own in London with his previous victory coming in the 4x200 freestyle relay.

"Going into every call room, I said it's my last semifinal or my last prelim or my last semi of the 100 fly, so tonight is the last semi ever," said Phelps, who plans to retire from swimming as soon as he touches the wall for the final time in London. "We're kind of chalking up all the lasts of certain things."

Lochte shook hands with his rival before crawling out of the pool for the last time at these games. In a symbolic gesture, he tossed his cap and goggles into the crowd, his work done. His final tally: two golds, two silvers, one bronze and a fourth-place finish ? impressive, but undoubtedly shy of what he had predicted would be "my time."

This time still belongs the Phelps.

At least for a couple more days.

"Ryan has probably been one of the toughest competitors I've swam against, all-around competitors," Phelps said. "We're seeing a lot more competitors coming up."

Rebecca Soni made quite a splash, too, on a night dominated by the Phelps-Lochte showdown.

Tearing through the water in her favorite pink suit, Soni set her second world record in as many days to defend her Olympic title in the 200 breaststroke. She finished in 2:19.59, breaking her own mark of 2:20.00 set in the semifinals.

Soni broke into a big smile when she saw the time, racing the clock more than she was anyone in the water. Japan's Satomi Suzuki took silver, more than a second behind at 2:20.72, while Russia's Yulia Efimova claimed bronze in 2:20.92.

"I'm so happy," Soni said. "I can't believe I did it."

Ranomi Kromowidjojo carried on the Dutch tradition of producing top sprinters and prevented a red, white and blue sweep of the night, taking the 100 freestyle in an Olympic-record 53.00. Aliaksandra Herasimenia of Belarus claimed the silver in 53.38, while the bronze went to China's Tang Yi in 53.44.

American teenager Missy Franklin got off to a terrible start ? she was last at the turn ? and couldn't rally. She finished fifth, two-tenths off the podium. The other U.S. swimmer, Jessica Hardy, finished last in the eight-woman field.

"I'm really unsatisfied about the time, but a gold medal is a gold medal," Kromowidjojo said. "I'm really happy with the gold medal."

In Soni's victory, South Africa's Suzaan van Biljon led at the first turn, but the American quickly seized control on the second lap. She was comfortably ahead by the second turn, then turned on the speed for the record.

"It's been my goal since I was a little kid to go under 2:20," Soni said. "That's when my coach told me you're going to be the first woman to go under 2:19. I've been chasing it ever since. I'm just so happy."

While Lochte couldn't hold on in the backstroke, it was still quite a night for the Americans. Clary rallied on the final lap to pull off the upset in an Olympic-record 1:53.41. Japan's Ryosuke Irie also got by Lochte on the final stroke, taking silver in 1:53.78. Lochte's time was 1:53.94.

"You always have big dreams in your head that you think you might be able to pull off something like that," Clary said. "The fact that it just came to fruition is something that hasn't even processed in my mind yet. The fact that I'm now an Olympic champion and Olympic-record holder is something that is very humbling. It's also very motivating for the next four years."

Last year, Lochte looked as though he had surpassed Phelps at the top of the swimming world when he captured five gold medals at the world championships. The Floridian didn't come close to that total at the Olympics, failing to defend his Olympic title in the 200 back and coming up short of Phelps again in the 200 IM.

Lochte won his first race of the Olympics with a dominating performance in the 400 IM on the opening night of swimming, but that was his biggest highlight. He failed to hold on in the anchor leg of the 4x100 freestyle relay, leaving the Americans with a silver, and he finished off the podium in the 200 free.

He did pick up a relay gold in the 4x200 free.

Snoop Dogg wants you to know that he?s tired of hip-hop, is Bob Marley? reincarnated and is embracing reggae instead of the culture of guns he once? rapped about.

Also, he?s got a new name: Snoop Lion.

The artist said at a news conference Monday in New York that he was ?born? again? during a visit to Jamaica in February and is ready to make music that his? ?kids and grandparents can listen to.?

The former gangster rapper is releasing a reggae album called ?Reincarnated?? in the fall. He said that in Jamaica, he connected with Bob Marley?s spirit and? is now ?Bob Marley reincarnated.?

Bob Marley?s son Rohan attended the conference and gave Snoop his? blessing.

?I feel like I?ve always been Rastafarian,? Snoop said of the spiritual? Jamaican movement. While there, he said, he visited a temple, was renamed Snoop? Lion and was also given the Ethiopian name Berhane, meaning ?light of the? world.?

Snoop didn?t explain why he was switching from ?Dogg? to ?Lion,? but it?s? likely a reference to the Lion of Judah, a religious symbol popular in? Rastafarian and Ethiopian culture.

Later, he played five songs for a small crowd, including one called ?No Guns? Allowed.? It features his daughter and includes the lyrics, ?No guns allowed in? here tonight, we?re going to have a free for all, no fights.?

?It?s so tragic that people are doing stupid things with guns,? he said.

Snoop, best known for hits like ?Gin and Juice? and ?Drop It Like It?s Hot,?? is an avid supporter of marijuana rights and has been banned from entering? Norway for two years after trying to enter the country with a small amount last? month.

He said that in Jamaica, where he stayed for 35 days, he grew closer to his? wife, who saw his transition. He added that he?s excited to perform music that? his family and children can listen to.

?As a 40-year-old man ? I?ve got to give them something,? he said. ?That?s? what you do when you?re wise.?

Snoop Dogg said he?s not completely retiring from hip-hop but is ?tired? of? the genre because it is no longer challenging.

?Reggae was calling ? it?s a breath of fresh air,? he said. ?Rap isn?t? challenging; it?s not appealing.?

The album was produced by Diplo and will feature Snoop singing. It will be? released on Vice Records.

The album will be followed with a documentary of the same name, also produced? by Vice. It features Snoop making music and will include some personal elements? of his life, a producer of the film said. It will debut at the Toronto? International Film Festival in September.

A coffee table book about Snoop?s rebirth is also in the works.

?It feels like I?m 19 or 20 years old again,? he said.

    Schmidt raised no donations for legal expense account in 2nd QTR ...

    Rep. Jean Schmidt, R-Miami Township, did not raise any reportable contributions for her legal expense trust from May through June, according to her latest filing with the House ethics committee.

    Schmidt set up the trust last year, after the ethics panel ordered her to repay about $500,000 in improper legal fees she had unknowingly received from the Turkish Coalition of America. The ethics committee said she could use donations raised through the trust to repay most of her legal debts.

    But so far this year, Schmidt has only raised one donation?a $5,000 contribution from Global Eclipse, a Delaware limited liability company with some Turkish ties, which donated to her in January. At the end of 2011, she still owed at least $515,000 in legal fees, according to her most recent financial disclosure form.

    If Schmidt isn?t pushing hard to raise money for her legal expense trust, it?s no wonder. She lost the GOP primary election in March to Brad Wenstrup, who will face William R. Smith in the general election. And Schmidt can disregard the ethics committee ruling once she leaves the U.S. House, according to ethics experts. Her term will be up in January 2013.

    (AP) ? Mysterious crop circles have appeared in an eastern Washington wheat field ? not far from the nation's largest hydropower producer ? but area farmers preparing for the summer's harvest find the distraction more amusing than alarming.

    "You can't do anything other than laugh about it," said Cindy Geib, who owns the field along with her husband, Greg. "You just kind of roll with the theory it's aliens and you're special because aliens chose your spot."

    Friends called the Geibs on July 24 when the pattern of flattened wheat was spotted off Highway 174, about five miles north of the town of Wilbur. The field is about 10 miles south of the Grand Coulee dam, which the Bureau of Reclamation says is the largest hydropower producer in the United States.

    The circles resemble a four-leaf clover and remind Cindy Geib of Mickey Mouse ears. The design knocked down about an acre of their wheat. Some of it could be salvaged by combines when the harvest starts in a week or two, she said, but some will be lost.

    "Of course, we don't have alien insurance," she said.

    Crop circles have been a worldwide phenomenon for decades, and this is not the first one in Lincoln County. Similar circular patterns were left in crops in the Wilbur area in 2010 and in 2008 or 2009, Geib said.

    Lynne Brougher, public affairs officer for the Grand Coulee dam, hadn't heard about the latest crop circles but said the previous one was no cause for alarm.

    "It seemed to be highly unusual," Brougher said. "As I recall from a couple of years ago, there was no good explanation of how they got there."

    Still, she added, "it wasn't a concern."

    The latest crop circle was first reported Tuesday by Spokane station KHQ-TV (http://is.gd/57FTpy ). There were no signs that anyone walked into the field.

    "We're trying to figure out how they got out there without breaking any of the wheat. It's hard to walk through the crunchy wheat and not knock it down," Geib said. "At the same time, it's hard to think it's aliens. It's a bizarre thing to wrap your brain around."

    Geib's daughter-in-law, Kelly Geib of Wilbur, says the crop circle has given the family something to ponder and chuckle about.

    "The kids all like to say the aliens have come, and we're happy to indulge them," she said.
